Powering on the SLICE System
Overview

To capture images of a sample, turn on both the SLICE microscope and BrightSLICE software.

To process or visualize images that were acquired previously, launch BrightSLICE software. The SLICE microscope should be off (powered down).

Powering on SLICE and BrightSLICE for microscopy and image acquisition

Turn on the computer that runs the SLICE microscope and log in.

Power on the SLICE microscope by pushing the main power switch on the back of the instrument.
When on, the SLICE power toggle will be illuminated in green and the small, red, camera-power indicator light will be on.

Verify that there is no sample inside the SLICE microscope and that the Refractive Index (RI) Media Reservoir is clear of obstructions.
Open the door and make sure that the SLICE stage is empty; no cuvette should be mounted.
This is important because, if a cuvette were attached to the stage during SLICE system stage initialization that launches upon startup, it would likely damage the microscope.

Launch BrightSLICE software and choose the User Profile that corresponds with the magnifying power of the system (i.e., 10x or 5x):
In the Select User Profile window that opens:

Check the Group that is selected and update it if needed.

Choose the Profile associated with the installed detection objective (SLICE 10x/5x) or a new profile based on this default profile.
The default SLICE 10x or SLICE 5x profiles display panels for controlling the microscope, such as the Software Joystick panel and the SLICE Light Sheet panel.

Click in the BrightSLICE stage initialization window that opens.
The stage will move through its travel range and return to the upper load position.
The window will close with no errors reported to indicate that initialization completed successfully.
When stage initialization is complete, you can load a sample and begin your work on the SLICE system.
Launching BrightSLICE software only for viewing and processing images

Turn on the computer that runs the SLICE microscope.

Launch BrightSLICE software and choose the User Profile that corresponds with the magnifying power of the system (i.e., 10x or 5x):
In the Select User Profile window that opens:

Check the Group that is selected and update it if needed.

Choose the Profile called Visualize & Process.
The Visualize & Process profile does not display microscope control panels, leaving more display space to view and process images that were acquired previously.
