Tracing from a multi color-channel image in 3D

Before you start tracing in the 3D window, you may want to select a single color channel for tracing (note that this is required for puncta detection).

  1. In the 3D environment, click Open Image to load a multichannel image.
  2. If desired you can associate traced structures with individual color channels by selecting the color channel for display using either the Channel panel on the left side of the 3D Environment window or the Image Adjustment tool (on the Image and Workspace ribbons) in the 2D window.
  3. (if needed) In the 2D window, use the Image Adjustment panel to make adjustments to visualize the structures you want to trace.
  4. Click Tree, Vessel, or Soma to start tracing.
