Acquire Time Sequence

Use for timed acquisitions, instead of Acquire Image or Acquire Stack.

Options are available to specify the type of acquisition (single, stack, multichannel)

1. Click the Acquire Time Sequence button ; the Acquire Sequence dialog box opens .
2. In the dialog box, select the appropriate lens and device/state pair. See Device Command Sequences to learn about device/state pairs.
3. Under Timer Settings, select 2D or 3D and set up the time settings.
   You selected "3D + time"

   To adjust the stack settings, click 4D Settings; the Stack Settings dialog box opens .

   • Top/Bottom Settings (automatic): Collect from top to bottom.
   • Area of Interest Focus (manual method): Microlucida prompts you to focus on the area of interest.
   • Interval: Amount of time between each acquisition.

   If the interval is less than the camera setting, you may lose images. Anything below .5 seconds may cause problems. a minimum setting of 1 second is best. Remember, after acquiring an image, the system may take 100 milliseconds or longer to process it. If the system is busy when a scheduled acquire interval "hits", the acquire does not happen—they system waits until the next scheduled acquire time.

4. Under Store Acquired Images, set the location, size, and format of your acquired images.
   • Size: Expressed in pixels. Camera Binning: not working?
5. Click Acquire Sequence.
   You selected "Area of Interest Focus" in the "4D Settings"

   1. The Focus dialog box opens . Focus then click OK.
   Change Hardware Settings
   Initial hardware settings are defined in Hardware Settings.
   The button leads to the Device Control dialog box .

   • Run Device Command Sequence & Change the State of a Device - Device Command Sequences

   1. The Image Stack Acquisition dialog box opens . Make the necessary adjustments and click OK.
      • To use Multichannel Acquire, your system must already be set up for a multichannel acquisition.
      • Use Change Hardware Settings/Multichannel Control as described above (step 5.a)
